Just want to ask if it is possible to make it mandatory that all Sales Order will originate from Sales Quotation in AX 2012 R2?

Should be a customization I believe.
You could try removing direct new creation on the sales table through security, but in reality you need to have the business process in place to ensure the starting point is always a quote, this is less of a system requirement and more of adherence to business processes.

If we want to follow the sales order life cycle in ax 2012 first we need to create a quotation and then sales order. Directly if we want to originate all sales orders from sales quotation we need to customize accordingly
