I have added a Wait and Move time to the route for a work centre but have noticed that this has no effect in the calculation of the Production Lead Time. Please can you confirm where I can see the effect of these fields.

It does have effect on production order lines. But the values are not copied directly from work center card. When you create a routing, default times are copied into the routing line, and then into production order when you refresh it.
If you recreate the routing line that uses this machine center and then refresh the production order, you’ll see the effect.
Also check your routing group settings for timing types to be included.
